Antioxidant-oligonucleotide conjugation refers to the covalent attachment of antioxidant molecules to oligonucleotides, which is a binding strategy that combines the therapeutic potential of oligonucleotides with the antioxidant properties of specific molecules and aims to enhance the stability and efficacy of oligonucleotide-based therapies. Antioxidants scavenge reactive oxygen species (ROS) and prevent oxidative damage to molecules that can reduce the oxidative stress that can occur during oligonucleotide delivery, cellular uptake and intracellular processing. However, oxidative stress is one of the major causes of oligonucleotide degradation, inactivation and off-targeting.
Bifunctional junctions with antioxidant and oligonucleotide reaction groups are used, which allow selective binding of the antioxidant to specific sites on the oligonucleotide molecule. The choice of antioxidant depends on stability, ROS scavenging ability and compatibility with conjugation chemistry to meet the needs of research.
