Introduction

The emergence of gene synthesis and targeted mutagenesis has driven the need to synthesize long oligonucleotides (>75 nucleotides). While there is a high demand for long oligonucleotides (150 mer and more), the yields and associated side reactions of conventional DNA synthesis previously limited the availability of oligonucleotides to<150 nucleotides. The longer the oligonucleotide, the more difficult it is to isolate the pure bioactive product. No chemical reaction can be completed with 100% efficiency. In oligonucleotide synthesis reactions, conjugation, capping, oxidation and deblocking, any of these defects can lead to faulty sequences. The more the accumulation of failed sequences, the lower the possibility of obtaining pure oligonucleotides. This resulted in a mixture of n-1 deletion mutations.
