IRES-EGFP IVT circRNA

IRES-EGFP IVT circRNA is a circular RNA designed for efficient and stable expression of enhanced green fluorescent protein (eGFP). This construct incorporates an Internal Ribosome Entry Site (IRES) element, which enables cap-independent translation initiation, making it particularly useful for applications where traditional linear mRNA may be less stable or less efficient. The circular structure of the RNA provides increased stability and resistance to degradation compared to linear mRNA, resulting in prolonged protein expression. The IRES-EGFP IVT circRNA is designed with optimized 5' and 3' untranslated regions (UTRs) and a poly(A) tail to further enhance translation efficiency and stability. Research has shown that incorporating specific RNA-binding protein motifs, such as those for poly(A)-binding proteins (PABPs), can significantly improve translation efficiency. Additionally, the use of engineered IRES elements, such as those with eIF4G-binding aptamers, can further enhance translation. This circRNA has been validated in various cell types, demonstrating robust eGFP expression and prolonged protein production compared to linear mRNA. Its enhanced stability and translation efficiency make it an ideal tool for applications requiring long-term expression of eGFP, such as in gene expression studies, cell tracking, and in vivo imaging experiments.
